Description We focused on the ribo-circRNAs circMbl, circCdi, and circPde8 and used as negative controls circRNAs for which we did not observe RFP reads in this cell type: circHaspin and circCamK1. We engineered the minigenes to express a V5-tagged protein in case of a circularization event (Figure 2A). Cells transfected with circMblV5, circCdiV5, or circPde8V5 minigenes produced V5-tagged proteins, whereas those transfected with circHaspinV5 or circCamK1V5 minigenes failed to express a V5-tagged protein. we found a consistent and very high confidence hit for a peptide that can only be produced by circMbl3. This circRNA encodes a 37.04 kDa protein and might either include or skip an alternative internal exon.
